REPOTMNF-κB報告基因主要應(yīng)用于NF-κB信號通路的研究和基于NF-κB信號通路的藥物研究以及癌細胞中NF-KB的調(diào)控機制的研究,。
pGMNF-ΚB-Lu是吉滿生物改造后的哺乳動物真核表達載體,,在其多克隆位點插入了多個NFκB結(jié)合位點,可以高靈敏度地檢測NFκB的激活水平,。同時,,對載體中預(yù)測出的其它轉(zhuǎn)錄因子以外的結(jié)合位點進行了適當?shù)耐蛔儯黾恿速|(zhì)粒的轉(zhuǎn)錄因子結(jié)合特異性,。另外,,由于質(zhì)粒體積減小,使得NFκB報告基因質(zhì)粒更易于轉(zhuǎn)染,。

pGMNF-Κb-Lu可以采用常規(guī)轉(zhuǎn)染方法轉(zhuǎn)染哺乳動物細胞,。用熒光素酶檢測試劑盒或雙熒光素酶檢測試劑盒進行檢測。
TNF-α,、IL-1β和LPS等是常見的NF-κB激活劑,,可作為NF-Κb報告基因的陽性對照,。
