•  bace1（β-site app cleaving enzyme）,，即β-分泌酶,，是淀粉樣前體蛋白（amyloid precursor protein，app）產(chǎn)生aβ的關(guān)鍵酶,。bace1屬于ⅰ型跨膜蛋白,，由501個(gè)氨基酸組成，其中46～460位氨基酸為功能活性區(qū)域,。bace1通過(guò)水解app670met-671asp之間的肽鍵,，生成670個(gè)氨基酸的可溶性n末端和包含完整aβ區(qū)的app跨膜c端。后者經(jīng)γ分泌酶進(jìn)一步酶切形成aβ,。zui近的研究表明,，bace1基因敲除小鼠腦內(nèi)的aβ水平可降低90%，且其生理表型基本正常,，顯示了bace1抑制劑在ad治療中的廣闊前景［1］,。通過(guò)酵母或昆蟲表達(dá)系統(tǒng)表達(dá)活性bace1在zui近已見(jiàn)報(bào)道［2，3］,。然而,，由于上述系統(tǒng)表達(dá)的蛋白量低，且純化流程復(fù)雜,，不適合bace1抑制劑的大規(guī)模篩選,。而bace1的原核表達(dá)可克服以上缺陷，為篩選特異性抑制劑及研究bace1結(jié)構(gòu)與功能關(guān)系提供大量材料,，為zui終從根本上治療ad帶來(lái)希望,。
