In this paper, the method of Alexa Fluor 647 (AF647) infrared fluorescein labeling (AAV2) was established, and the structure and morphology of the labeled virus were compared with those of the unlabeled virus by silver staining, infrared fluorescence imaging and transmission electron microscope analysis of gel electrophoresis. At the same time, its transduction activity after infection with 293T cells was further identified, The physicochemical properties and infective activity of AAV2 remained unchanged after labeling. The cell uptake efficiency and distribution of the labeled virus in host cells were described using slit imprinting and laser confocal microscopy imaging techniques, providing an example for the application of infrared labeled AAV2 in research AF647 labeled AAV2 provides a new means for studying the infection kinetics of adeno-associated viruses, with advantages such as high sensitivity, anti fluorescence bleaching, and low non-specific interference. It also has important application value for directly and effectively combining morphological analysis and biochemical molecular biology detection.
