熱熔擠出技術(shù)(Hot-Melt Extrusion technique,,HME)又可稱為熔融擠出技術(shù)(Melt Extrusion technique),,是近年來歐洲、北美和日本大力開發(fā)的一種新的制劑技術(shù),,主要用于提高難溶性藥物的溶出度,,制備緩控釋制劑等。

熔擠出的用途
l 可用口感不好藥物掩味工藝,,將藥物與掩味的高分子材料一起熱融制粒后粉碎,,可很好掩味,為進(jìn)一步制備口服液,、干混懸劑或口崩片奠定良好基礎(chǔ),。
l 可直接制備緩釋顆粒,將藥物與高分子緩釋材料一起熱融制粒,,可一步制得十分均勻 的緩釋顆粒,,可制成其它緩釋制劑。

擠出原理
l HME技術(shù)是將2種或2種以上的物料加入到逐段控溫的機筒中,機筒內(nèi)設(shè)置螺桿元件,,螺桿元件從加料部位到機頭順次執(zhí)行不同的單元操作,,物料在螺桿的推進(jìn)下前移,在一定的區(qū)段熔融或軟化,,熔體在剪切元件,,混合元件的作用下均勻混合,,zui后以一定的壓力,,速度和形狀從機頭口模擠出。
l HME技術(shù)的基本思想是在一個軸向空間內(nèi)連續(xù)設(shè)置多種單元操作,,使多組分物料在經(jīng)由這一空間的過程中粒徑不斷減小,,同時彼此間進(jìn)行空間位置的對稱交換和滲透,zui終達(dá)到分子水平的混合,。
