Cytokeratin 17(細(xì)胞角蛋白17)

細(xì)胞角蛋白17 分子量為46kDa,,表達(dá)于復(fù)層上皮和氣管、喉,、支氣管假?gòu)?fù)層柱狀上皮的基底細(xì)胞以及唾液腺的肌上皮細(xì)胞,。在腫瘤組織中,該抗體主要識(shí)別肺,、子宮頸和口腔的鱗狀細(xì)胞癌,。主要用于鱗狀細(xì)胞癌的研究。
診斷應(yīng)用:

1. 滑膜肉瘤
2. 上皮樣肉瘤
3. 肉瘤中的異常染色可為間皮瘤,、黑色素瘤,、平滑肌肉瘤和內(nèi)皮細(xì)胞腫瘤的診斷提供線索,通常只有CK8和CK18(被Cam5.2和35BH11證明)表達(dá)陽性,,這兩種存在于所有組織中的原始角蛋白胚胎源性表達(dá)也許可以解釋CK8和CK18的異位性表達(dá),。
4. 在小細(xì)胞癌中可以看到細(xì)胞核周“圓點(diǎn)狀”染色,如美克爾細(xì)胞癌,、肉瘤及髓細(xì)胞瘤可顯示異常免疫反應(yīng)性,。
5. 許多“兒童的小藍(lán)圓細(xì)胞腫瘤”表達(dá)角蛋白,包括尤文氏肉瘤/PNET,、橫紋肌肉瘤,、腎母細(xì)胞瘤和促結(jié)締組織增生性小圓細(xì)胞腫瘤,。

除了表格所列的20種軟角蛋白外,還有限于頭發(fā),、指甲上皮組織的硬角蛋白,。細(xì)胞角蛋白zui初的分類建立在2D凝膠電泳的基礎(chǔ)上。現(xiàn)在有特異的單克隆抗體可應(yīng)用于石蠟包埋的組織,,由于細(xì)胞角蛋白之間的同源性,,單抗需十分仔細(xì)地篩選以避免交叉反應(yīng)??乖瓫Q定簇在某些情況下可以被遮蔽,,可能反應(yīng)發(fā)生在免疫印跡上而不是在組織切片,。

Cytokeratin 17 has a molecular weight of 46 kDa and is expressed on the epithelium of the stratified epithelium and in the trachea, larynx, bronchial pseudostratified columnar epithelium, and myoepithelial cells of the salivary gland. In tumor tissue, the antibody mainly recognizes squamous cell carcinomas of the lung, cervix and mouth. Mainly used for squamous cell carcinoma research.
Diagnostic Applications:

3. Abnormal staining in sarcomas can provide clues to the diagnosis of mesothelioma, melanoma, leiomyosarcoma and endothelial cell tumors, which are usually only positive for CK8 and CK18 (evidenced by Cam5.2 and 35BH11), both present in all tissues The original keratin-derived expression may explain the ectopic expression of CK8 and CK18.
4. Small cell carcinoma can be seen in the perinuclear "dot-like" staining, such as melanocarcinoma, sarcoma and myeloblastoma can show abnormal immunoreactivity.
5. Many "small blue circle cell tumors in children" express keratin, including Ewing's Sarcoma / PNET, rhabdomyosarcoma, Wilms' tumor, and connective tissue proliferating small round cell tumors.
Cytokeratin Moll number
In addition to the 20 types of soft keratins listed in the table, there is limited to keratin hair and nail epithelium. The initial classification of cytokeratin was based on 2D gel electrophoresis. Specific monoclonal antibodies are now available for use in paraffin-embedded tissues. Due to the homology between cytokeratins, the monoclonal antibodies should be carefully screened to avoid cross-reactivity. Epitopes can be obscured in some cases, and the reaction may occur on immunoblots rather than on tissue sections.
