Enzo Life Sciences的CYTO-ID®綠色/紅色長期細胞示蹤試劑盒采用專有的非共價細胞標(biāo)記技術(shù),,將含有疏水性脂肪鏈的熒光染料穩(wěn)定地結(jié)合入細胞膜的脂質(zhì)雙分子層中。標(biāo)記緩沖液對哺乳動物細胞是等滲的,,不含去污劑或有機溶劑,。染料對細胞沒有毒性,可在細胞中保留長達96小時,,并在有絲分裂時傳遞給子細胞,。由于該染料不對細胞內(nèi)的蛋白質(zhì)進行共價修飾,與基于硫醇反應(yīng)的氯甲基或胺反應(yīng)的琥珀酰亞胺酯類熒光染料的分子探針相比,,細胞正常的生理反應(yīng)得到更好的保留,。
也可以使用CELLESTIAL®染料進行雙重標(biāo)記。標(biāo)記的細胞可以通過共聚焦熒光顯微鏡等儀器進行觀察,。此外,,染料標(biāo)記的和未標(biāo)記的細胞群可通過流式細胞儀進行分析。在延長96小時的培養(yǎng)后,,沒有觀察到熒光轉(zhuǎn)移到相鄰細胞,。這與Calcein AM和BCECF AM標(biāo)記形成鮮明對比,后者在生理溫度下只能在存活的細胞內(nèi)保留幾個小時,。
