| I. Background | |
| DLL4（Delta-like ligand 4） 主要表達于血管內皮細胞,，通過 “細胞-莖細胞"分 化機制調控血管分支：DLL4 激活鄰近細胞的 Notch1 信號,，抑制其分化為細胞,，從而 限制血管過度生長，維持血管網絡的有序性,。在腫瘤微環(huán)境中,，DLL4 在異常血管中高表 達，誘導血管結構紊亂（滲漏,、低灌注）,，促進缺氧和耐藥。此外，DLL4 參與神經 發(fā)育及 T 細胞分化,，其基因突變可導致 Adams-Oliver 綜合征等先天性疾病,。 DLL4 與 Notch1 是 Notch 信號通路 的關鍵組分，通過細胞間接觸依賴性信號調控發(fā)育,、 血管生成及疾病進程,。DLL4 是 Notch 配體家族成員，作為跨膜蛋白特異性地結合并激 活 Notch1 受體,，驅動細胞命運決定,、組織穩(wěn)態(tài)及病理重塑。 | |
