| I. Background | |
血小板衍生生長因子（PDGF）是一類重要的生長因子,，主要由血小板,、內(nèi)皮細(xì)胞和 巨噬細(xì)胞分泌，在細(xì)胞增殖,、遷移,、分化和血管生成等生物學(xué)過程中發(fā)揮關(guān)鍵作用,。它不 僅在正常生理過程中（如傷口愈合和組織修復(fù)）起重要作用,，還在多種病理過程中（如癌 癥、纖維化和動(dòng)脈粥樣硬化）扮演重要角色,。PDGF 家族包括 PDGF-A,、PDGF-B、PDGF- C 和 PDGF-D 四種亞型,。PDGF 通過與其受體（PDGFR）結(jié)合,，激活下游信號通路，調(diào)控 細(xì)胞行為,。 血小板衍生生長因子受 0 體α（PDGFRA）,，又稱為 CD140A 或 PDGFR2，是一種細(xì)胞 表面激酶受體,，屬于血小板衍生生長因子家族。PDGFRA 與配體 PDGF 結(jié)合后會(huì)二 聚化以及磷酸化,，激活包括 JAK-STAT3,、PI3K-AKT-mTOR 和 RAS-MAPK 等下游信號通路， 在調(diào)節(jié)關(guān)鍵細(xì)胞功能如增殖和凋亡方面有重要作用,。PDGFRA 蛋白被激活異常時(shí),，則會(huì)導(dǎo) 致腫瘤的發(fā)生并促進(jìn)腫瘤血管生成,。 | |
