LYMPHOCYTE SEPARATION MEDIUM
LSM的優(yōu)勢(shì)：
傳統(tǒng)的組合是Ficoll+泛影酸葡甲胺（甲泛影鈉）,。Yeasen公司的LSM主要成分是Ficoll （聚蔗糖）和sodium diatrizoate （泛影酸鈉，密度1.200±0.002g/ml），每100ml包含6.2g聚蔗糖和9.4g泛影酸鈉,。目前為止,，這個(gè)組合是淋巴細(xì)胞分離液成分使用zui廣泛,、分離效果的組合,。
與LSM競(jìng)爭(zhēng)zui大的產(chǎn)品主要是：GE公司的Ficoll-Paque PREMIUM,，其特點(diǎn)如下：

使用說(shuō)明（用于去纖維蛋白或抗凝血?jiǎng)┨幚磉^(guò)的人體血液,；對(duì)于其他物種或者其他組織的血液，需要對(duì)此程序做出一定的改進(jìn)）
1. 輕輕顛倒瓶子使LSM充分混合
2. 無(wú)菌轉(zhuǎn)移3 mlLSM到15 ml離心管中,。
3. 混合2 ml除纖維蛋白血液或肝素處理血液 和2 ml 生理鹽水
4. 仔細(xì)的將稀釋血液加入3 mlLSM（室溫）于15ml離心管中,，在血液和LSM中形成一個(gè)明顯的分層。不要將稀釋血液混合入LSM中,。
5. 室溫下400g離心15-30分鐘,，離心可以沉淀紅細(xì)胞和多核白細(xì)胞同時(shí)可以再LSM上形成一層單核淋巴細(xì)胞，如下圖所示,。
6. 吸出淋巴細(xì)胞上方2-3mm的血漿,。
7. 吸取淋巴細(xì)胞層以及它下面一半的LSM和轉(zhuǎn)移到另外的一個(gè)離心管。加入等體積的平衡鹽緩沖液至淋巴細(xì)胞離心管中,，室溫離心10分鐘,，離心速度設(shè)定在既不損傷細(xì)胞又能沉淀細(xì)胞即刻，例如160 - 260 x g（去除LSM和降低血小板的百分比）,。
8. 用平衡鹽緩沖液清洗細(xì)胞,，用適當(dāng)?shù)呐囵B(yǎng)基重懸細(xì)胞。
