酶聯(lián)免疫吸附試驗(yàn)
       ①直接法    該方法直接利用抗原 與酶聯(lián)抗體的結(jié)合，較簡(jiǎn)單易行,。A.假抗原,。在聚苯乙烯酶標(biāo)版的孔穴中加入待檢測(cè)的抗原（捕食者胃內(nèi)含物抽提液）0.2mL，在4℃冰箱中過(guò)夜,，使抗原黏附于塑料孔壁上,。B.洗滌兩次，倒置于干毛巾上,，使孔穴中的液體盡量流出,。c.加酶聯(lián)抗體。 各孔穴中加入酶聯(lián)抗體0.2mL,，在mL37℃下孵育3h,，使其充分結(jié)合。d. 加底物,。 用PBS充分洗滌以除去未結(jié)合的酶聯(lián)抗體,，然后加入底物（鄰苯二胺）0.2 mL，于37℃反應(yīng)30min,，用2mol/L硫酸0.05 mL終止反應(yīng),。e. 用ELISA檢測(cè)儀測(cè)定其吸光度值，分別設(shè)陽(yáng)性,、陰性和空白對(duì)照,，以確定陽(yáng)性反應(yīng)臨界值。
        ②雙抗體夾心法   該方法與直接法的不同在于吸附抗體,。a. 包被抗體,。酶標(biāo)板的孔穴中加入0.2 mL抗體（用PH=9.6/0.05mol/L碳酸鹽緩沖液稀釋?zhuān)?7℃水浴中孵育3h,移入4℃冰箱過(guò)夜,。將各孔中多余的抗體傾去,，加入PBS洗滌，立即甩去,，再將PBS加滿各板孔,，置5min,甩去溶液，如此洗滌2次,，倒置于干毛巾上,，使孔穴中的液體盡量流出。b. 加待檢抗原,。沒(méi)孔中加入待檢捕食者胃內(nèi)含物抽提液0.2 mL.同時(shí)設(shè)陽(yáng)性,、陰性和空白（PBS）對(duì)照，37℃水溶中孵育3h,，轉(zhuǎn)入冰箱中過(guò)夜,。如上法洗滌3次。C. 加入酶聯(lián)抗體,。每孔加入酶聯(lián)抗體0.2 mL,，37℃水浴孵育2h,，如上法洗滌3次。d. 加底物,。 每孔加入底物鄰苯二胺0.2 mL, 37℃水浴孵育1h,加入2mol/L硫酸0.05,，以終止反應(yīng)。e. 用 ELISA檢測(cè)儀測(cè)定其吸光度值,。
       ③間接法  與前兩種方法不同點(diǎn)在于酶聯(lián)抗體與抗原的間接結(jié)合,。a. 每孔加入待檢抗原0.2 mL ，4℃冰箱中過(guò)夜,，使抗原黏附于塑料孔壁上,。并依上法洗滌3次。b. 每孔加0.2 mL,用PBS稀釋的抗體,，37℃孵育2-3h,。c.用PBS洗滌3次后，加入溶于PBS的酶聯(lián)抗體0.2mL,，37℃孵育2-3h。d. 加底物,，方法同上,。e. 用ELISA檢測(cè).
