血清

使用血清分離管,，讓樣品在室溫下凝固2小時或在4℃下過夜，然后以約1000×g離心20分鐘,。立即測定新鮮制備的血清，或?qū)悠贩盅b在-20℃或-80℃下備用,。避免反復(fù)凍/融循環(huán),。

等離子體

使用EDTA或肝素作為抗凝血劑收集血漿。在收集后30分鐘內(nèi),，在2-8℃下以1000×g離心樣品15分鐘,。立即去除血漿并進行檢測，或?qū)悠繁４嬖?20℃或-80℃的等分試樣中以備后用,。避免反復(fù)凍/融循環(huán),。

組織勻漿

組織勻漿的制備將根據(jù)組織類型而變化。通常,，組織應(yīng)在冰冷的PBS（0.02mol / L,，pH 7.0-7.2）中*沖洗以除去過量的血液并在均質(zhì)化前稱重。將組織切成小塊并在冰上用玻璃勻漿器在5-10mL PBS中均化（Micro Tissue Grinders也起作用）,。應(yīng)使用超聲波細胞破碎器對所得懸浮液進行超聲處理,，或進行兩次凍融循環(huán)以進一步破壞細胞膜。之后,，勻漿應(yīng)以5000×g離心5分鐘,。立即取出上清液并進行分析，或等分試樣并在≤-20℃下儲存,。

細胞裂解物

在冷PBS中洗滌細胞三次,。

注意：

樣品在5天內(nèi)可以被存儲在4使用Ô C,，否則樣品必須儲存在-20 Ô C（≤1月）或-80 Ô C（≤2個月），以避免生物活性和/或污染的損失,。
樣品溶血會影響結(jié)果; 不會檢測到用作樣本的溶血標本,。

標準

傳統(tǒng)的ELISA試劑盒：使用前短暫旋轉(zhuǎn)或離心檢測A和檢測B溶液,。分別用Assay Diluent A和B稀釋至工作濃度（1：100）。

即用型ELISA試劑盒：檢測溶液A和B的濃度已經(jīng)正確,，無需進一步稀釋,。

洗液

用580mL去離子水或蒸餾水稀釋20mL洗滌液濃縮物（30×），制備600mL洗滌液（1×）,。

TMB底物

用滅菌的吸頭吸取所需劑量的溶液,。不要將殘留的溶液倒回到小瓶中。

注意：

不要直接在孔中進行連續(xù)稀釋,。
在進行測定后15分鐘內(nèi)準備標準品,。不要直接在37 o C 溶解試劑,。
檢測試劑A和B是粘性溶液，因此慢慢移液它們以減少體積誤差,。
傳統(tǒng)的ELISA試劑盒：根據(jù)說明小心地重新構(gòu)建標準品或使用檢測試劑A和B,，避免起泡并輕輕混合直至晶體*溶解。為了大限度地減少移液造成的不性,，請使用小體積并確保移液器已校準,。建議一次移液10μL以上以確保準確性。
即用型ELISA試劑盒：重組標準品只能使用一次,。

樣品制備

Reddot Biotech僅對試劑盒本身負責,，而不是對試驗過程中消耗的樣品負責。實驗者應(yīng)該在開始實驗之前總是計算所需的樣品總量,。
實驗者還應(yīng)該在開始之前預(yù)測大致的濃度,。如果預(yù)期值不在標準曲線范圍內(nèi)，建議確定樣品的宜稀釋度,。
如果您使用的樣品類型未在手冊中指出,，則應(yīng)在開始實際實驗之前進行初步實驗以確保試劑盒的有效性。
建議不要使用化學裂解緩沖液來制備組織或細胞提取樣品,，因為它可能會由于裂解緩沖液的含量而導(dǎo)致意外的反應(yīng)和結(jié)果,。
由于其他來源的抗原與我們試劑盒中使用的抗體之間可能存在錯配，因此不建議使用其他制造商生產(chǎn)的天然或重組蛋白,，因為我們的試劑盒可能無法識別它們,。
我們的試劑盒可能無法檢測到細胞培養(yǎng)上清液中的樣品，因為它們很容易受到細胞活力,，細胞數(shù)量和/或取樣時間等因素的影響,。
建議將未經(jīng)長期儲存的新鮮樣品與我們的試劑盒一起使用。如果樣品長時間未冷凍保存,，可能會發(fā)生蛋白質(zhì)降解和變性,，導(dǎo)致結(jié)果相互矛盾或不正確。

分析程序

用于夾心ELISA試劑盒

確定稀釋標準品,，空白樣品和樣品的孔,。為標準準備7口井，為空白準備1口井,。每種稀釋的標準品（讀取試劑制備）,，空白和樣品加入100μL到適當?shù)目字?。用Plate密封劑蓋住。在37 o C 孵育2小時,。
