KlenTaq-S酶——適PAP聚合酶

PAP(焦磷酸化激活的聚合反應(yīng)法)是一種*的DNA擴增技術(shù)（Liu and Sommer: BioTechniques, 29 [2000], 1072-1080; Nucleic Acids Res, 30 [2002], 598-604),該技術(shù)利用DNA聚合酶在3'封閉的引物進行連續(xù)的焦磷酸化和聚合反應(yīng),。焦磷酸化是DNA聚合的逆反應(yīng),，其引物模板雙鏈的3'末端封閉核苷酸（通常是雙脫氧）在無機焦磷酸鹽存在下被酶解清除，在焦磷酸化清除封閉的核苷酸后,，激活引物進行后續(xù)的DNA聚合,。由于PAP的高特異性（1 / 3*10¹¹），該技術(shù)正成為檢測存在大量野生型等位基因的情況下檢測稀有突變,，大雜合缺失和重復(fù)基因的有力工具（Liu et al：Biotechniques,， 40 [2006],，661-668）。理論上,，PAP可以檢測3*10¹¹個拷貝的野生型等位基因中的單堿基突變,，此外，PAP還可用作替代或證實real-time PCR反應(yīng) (Boon et al : Prenat Diagn 27[2007], 932-937),。

KlenTaq-S酶：

KlenTaq-S已被多個研究團隊證實是PAP反應(yīng)適的熱穩(wěn)定Taq DNA聚合酶,，該酶由5'缺失和F667Y（Tabor和Richardson）突變的基因表達,，編碼的Thermus aquaticus DNA聚合酶,。該酶反復(fù)暴露于98度的PR1反應(yīng)緩沖液中沒有表現(xiàn)出任何明顯的活性損失，具有*的熱穩(wěn)定性,。

KlenTaq-S酶的濃度 : 10 U/µl

建議PAP反應(yīng)條件 (25 µl反應(yīng)體系) :

50 mM Tris-HCl, 7.8,

16 mM 硫酸銨,

3.5 mM 氯化鎂,

25 µM each four dNTPs,

0.1 µM 引物,

5-200 ng模板,

90 µM 無機焦磷酸鹽 (Na4PPi),

2% DMSO,

循環(huán)條件 :

95度1 min (變性),

94度 15 sec

60度 30 sec

64度 30 sec

68度 1 min

72度 1-2 min

循環(huán)數(shù)25-45 ,。

緩沖體系 (10X PR1):

500 mM Tris-HCl, pH 7.8,

160 mM (NH4)2SO4,

35 mM MgCl2, 1.5 mg/ml BSA
