全自動(dòng)外泌體熒光檢測分析系統(tǒng)ExoView是美國NanoView Biosciences于2018年研究推出的,該系統(tǒng)經(jīng)推出,,便引起了外泌體域科研工作者的廣泛關(guān)注,。僅兩年已有100多個(gè)實(shí)驗(yàn)室采用該技術(shù),,發(fā)表文獻(xiàn)近百篇,刊載在Cell, Cell Biology, Journal of Extracellular Vesicles等期刊,。ExoView的精準(zhǔn)表征,,為科學(xué)家提供了種無須純化即可直接對(duì)血漿、血清,、尿液,、培養(yǎng)基中的外泌體進(jìn)行表征分析的新方法。能夠提供單個(gè)外泌體水平下的全·方位外泌體表征信息(包括粒徑大小,、分布,、計(jì)數(shù)、攜帶蛋白表達(dá)與生物標(biāo)志物(CD9,,CD81,,CD63等)共定位等),以及多層次和全方面的外泌體測量解決方案,。能夠幫助科學(xué)家更深入地了解外泌體與疾病之間的關(guān)系,,助力疾病診斷和新藥開發(fā)。
ExoView® R200全自動(dòng)外泌體熒光檢測分析系統(tǒng)勢包括:
• 異性捕獲:芯片上可包被多達(dá)6 種捕獲抗體,,異性捕獲含定蛋白標(biāo)記物的外泌體,。
• 對(duì)陽性外泌體計(jì)數(shù):芯片捕獲外泌體后,可通過單粒子干涉反射成像傳感器(SP-IRIS)技術(shù)直接檢測樣品中外泌體的數(shù)量,。
• 單個(gè)外泌體蛋白共定位分析:檢測每個(gè)外泌體的熒光信號(hào)并進(jìn)行統(tǒng)計(jì),,可獲得熒光共定位信息,用于分析樣品中不同表型外泌體的比例,。
• 無需純化:使用抗體捕獲模式,防止樣品中雜質(zhì)影響結(jié)果,,可直接檢測血液,、尿液和細(xì)胞培養(yǎng)液中的外泌體,未純化樣品的測量結(jié)果與純化后基本致,。
• 粒徑分辨率高:高精度SP-IRIS技術(shù),,可檢測≥50 nm的外泌體,測量結(jié)果與電子顯微鏡檢測結(jié)果基本致,,并統(tǒng)計(jì)生成外泌體的粒徑分布結(jié)果,。
• 可檢測外泌體內(nèi)容物:試劑盒配套相應(yīng)的穿膜劑,可穿透外泌體并對(duì)外泌體內(nèi)容物進(jìn)行染色并檢測,,如下圖所示,,Non-permeabilized時(shí)只能檢測到跨膜蛋白CD9的熒光信號(hào), Permeabilized后即可檢測到外泌體內(nèi)容物(如PD-L1,、P-tau等)的表達(dá),。
