Operation steps for cell culture
- 吸走用于培養(yǎng)基，加入3-5mL PBS后輕輕晃動(dòng)培養(yǎng)瓶潤(rùn)洗細(xì)胞,；
- 吸干凈PBS后加入1mL 0.25%-0.53mM EDTA,，輕輕搖晃培養(yǎng)皿使浸沒(méi)細(xì)胞表面，培養(yǎng)瓶放37度培養(yǎng)箱消化,；
（細(xì)胞對(duì)于消化比較敏感,，消化過(guò)度會(huì)嚴(yán)重影響細(xì)胞狀態(tài)，導(dǎo)致細(xì)胞傳代死亡漂浮或者傳代后不長(zhǎng),。細(xì)胞消化到細(xì)胞間隙變大但未脫落,，可以輕輕吹下時(shí)即可終止，禁止消化到細(xì)胞漂浮,?；靹蚣?xì)胞時(shí)盡量輕柔，不要吹出大量氣泡,。） - 加入6-8ml培養(yǎng)基終止消化,，輕輕吹下細(xì)胞混勻；
- 將混勻的細(xì)胞1000rpm（約150g）離心3min,，棄上清,，再用新鮮培養(yǎng)基重懸37度培養(yǎng)箱繼續(xù)培養(yǎng)；
